The Witcher 3 Blood and Wine: Envoys, Wineboys
This is the first main quest in Blood and Wine. It appears in the journal automatically after you install the expansion. One exception is when your character is at an early stage of the game and has not yet completed the main quest A Poet Under Pressure within the main storyline (which happens in Novigrad).
Important - Before you make a decision about accepting Anna Henrietta's invitation and setting off on a journey to Toussaint, make sure that Geralt is at least level 34. Otherwise, you can have a lot of trouble with the new quests available in the expansion as well as exploration of the duchy itself.

Visit any notice board in Velen and look for the announcement shown in the picture above, it will be personally addressed to Geralt.
Meet the knights from Toussaint in Holloway
The village you seek is located a short distance west of Oxenfurt. Namely the location marked as Stonecutter's Settlement on the map of Velen. After you reach the location, find Palmerin and Milton.
Help Palmerin and Milton defeat the bandits

the nearby village of Holloway under their protection, and plan to deal with the bandits plaguing its inhabitants. You can offer to help them in solving the bandit problem. Should you decide to do so, a brief cut-scene will be shown, after which the battle starts. Bandits have no unique skills, but you need to bear in mind that their experience level will be aptly matched to the expansion (around level 35).
Go with Palmerin and Milton to Toussaint
The last part of the quest can be approached in two ways - by defeating the bandits plaguing Holloway, or by picking the second dialogue option when you meet Palmerin and Milton for the first time. In the latter case, Geralt will dissuade the knights from taking the violent option.
Regardless of the circumstances, you will have to listen to the official invitation and learn of the mysterious beast murdering people in Toussaint. After Geralt expresses his readiness to accept the contract you will be transported to Toussaint and a new main quest - The Beast of Toussaint - will automatically start.
