Way to unlock the quest: Talk to Bones (at any point into the game).

You complete this mission during the conversation with Bones

It is a very short quest, for the completion of which you only need to talk to Bones for a while. You can do that at any point of the game, e.g. right after you meet with Bones for the first time (after he restores you at the Crab Coast). During the conversation, Bones raises the topic of the Isle of Thieves, which you need to ask him about.
